Can I lock 1 room and lease out my whole HDB house ?

My guess is that you have yet to fulfill MOP.
You can lock up, but you cannot term it as "leasing out whole HDB flat" because It is still considered as illegal as far as HDB is concern.
The legal way is to just rent out the rooms individually.

You can rent out rooms but not the whole unit if you have not meet the HDB MOP.
